目的:建立液相色谱柱前衍生法检测乳及乳制品中羟脯氨酸的方法。方法:样品经酸水解后,羟脯氨酸与异硫氰酸苯酯在碱性条件下衍生化,经反相液相色谱分离,二极管阵列检测器254 nm检出,外标法定量。结果:加标回收率在93.3%~104.6%之间,相对标准偏差为0.97%~4.3%。结论:方法准确、可靠,适合于乳与乳制品中是否添加动物胶原水解蛋白的鉴定。
Objective: To establish a method for the determination of hydroxyproline in milk and dairy products by pre-column derivatization. Methods: After acid hydrolysis, hydroxyproline and phenyl isothiocyanate were derivatized under basic conditions. The compounds were separated by reversed-phase liquid chromatography and detected by diode array detector at 254 nm. Results: The spiked recoveries ranged from 93.3% to 104.6% with relative standard deviations of 0.97% -4.3%. Conclusion: The method is accurate, reliable and suitable for the identification of animal collagen hydrolyzed proteins in milk and dairy products.
