通过研究超声波辅助酶法提取马尾藻多糖的最佳条件,为提高马尾藻的利用率提供理论依据。采用响应面分析优化酶解法提取多糖工艺与正交优化超声波破碎工艺,得到超声波辅助酶法提取多糖的最佳提取条件。结果表明:超声波辅助酶法提取马尾藻多糖最佳条件为:超声波作用时间40min,功率350W,工作温度80℃,酶解温度44.95℃、酶解pH5.0、酶解时间2.05h,酶添加量2.5%,所得马尾藻多糖提取率为17.43%,比单独酶解法提取马尾藻多糖提高了3.41%。
The optimal conditions for the extraction of polysaccharides from Sargassum by ultrasonic-assisted enzymatic extraction are provided to provide theoretical basis for improving the utilization rate of Sargassum. The polysaccharides extracted by the method of response surface analysis and optimized enzymolysis and the orthogonal optimization of the ultrasonic crushing process were used to obtain the optimal extraction conditions of polysaccharides by ultrasonic assisted enzymatic extraction. The results showed that the optimal conditions for ultrasonic extraction of polysaccharides from Sargassum were: ultrasonic time 40min, power 350W, working temperature 80 ℃, temperature 44.95 ℃, pH 5.0, hydrolysis time 2.05h, enzyme dosage 2.5%. The extraction rate of the resulting Sargassum polysaccharides was 17.43%, which was 3.41% higher than that of the polysaccharides extracted by the separate enzymatic method.
