目的分离并鉴定喉癌患者癌组织和癌旁组织的差异表达蛋白质,探讨喉癌发生、发展的分子机制。方法收集喉癌患者癌组织和癌旁组织标本各10例,二维电泳分离总蛋白质,鉴定有明显表达差异的蛋白质点,并对其进行质谱分析。结果获得了分辨率和重复性均良好的凝胶蛋白质图谱,筛选出46个有明显表达差异的蛋白质点,共有17个蛋白质点被成功鉴定,其中在喉癌组织中低表达的有5个,高表达的有12个。结论喉癌患者癌组织蛋白质的表达相对于癌旁组织存在明显差异,差异蛋白质的鉴定有望成为筛选喉癌特异性肿瘤标志物及药物治疗靶点的依据。
Objective To isolate and identify differentially expressed proteins in cancer tissues and adjacent tissues of laryngeal cancer so as to explore the molecular mechanism of the occurrence and development of laryngeal cancer. Methods Ten cases of laryngeal cancer tissues and adjacent tissues were collected. Two-dimensional electrophoresis was used to separate the total proteins. The protein spots with obvious differences were identified and analyzed by mass spectrometry. Results The gel protein profiles with good resolution and repeatability were obtained, and 46 protein spots with obvious expression differences were screened out. A total of 17 protein spots were successfully identified, among which 5 were low expressed in laryngeal carcinoma tissues, High expression of 12. Conclusion The expression of protein in cancer tissue of laryngeal cancer patients is obviously different from that of paracancerous tissues. The identification of differential proteins is expected to be the basis of screening specific tumor markers of laryngeal cancer and drug therapy target.
