目的:观察布地奈德对哮喘大鼠外周血中性粒细胞上糖皮质激素诱导的肿瘤坏死因子受体(GITR)及其配体(GITRL)表达水平的影响。方法:将30只清洁级SD大鼠随机分为3组:对照组、哮喘组、布地奈德干预组,取外周血进行中性粒细胞的分离提纯,采用实时荧光定量逆转录聚合酶链反应(QRT-PCR)方法检测GITR/GITRL mRNA在中性粒细胞上的表达。结果:哮喘组中性粒细胞上的GITR/GITRL mRNA的表达水平(△CT分别为3.10±0.23,13.48±2.40)显著高于对照组(△CT分别为5.16±0.70,15.79±1.27)及布地奈德干预组(△CT分别为4.83±0.93,15.63±1.73)(均P<0.05);而布地奈德干预组与对照组相比,差异无统计学意义(P>0.05)。对各组的GITR/GITRL mRNA表达水平进行直线相关分析,结果显示两者表达呈正相关(R2=0.669,P<0.01)。结论:中性粒细胞可能通过GITR/GITRL信号通路参与哮喘的气道炎症过程,布地奈德可能通过下调中性粒细胞上的GITR/GITRL的表达从而减轻哮喘气道炎症反应。
AIM: To observe the effect of budesonide on glucocorticoid-induced tumor necrosis factor receptor (GITR) and its ligand (GITRL) expression in peripheral blood neutrophils of asthmatic rats. Methods: Thirty clean-grade SD rats were randomly divided into three groups: control group, asthma group and budesonide intervention group. Peripheral blood was taken for neutrophil purification. Real-time fluorescence quantitative reverse transcription polymerase chain reaction (QRT-PCR) was used to detect the expression of GITR / GITRL mRNA on neutrophils. Results: The expression levels of GITR / GITRL mRNA in asthmatic neutrophils (△ CT were 3.10 ± 0.23 and 13.48 ± 2.40 respectively) were significantly higher than those in the control group (△ CT 5.16 ± 0.70 and 15.79 ± 1.27, respectively) Nerd intervention group (△ CT were 4.83 ± 0.93, 15.63 ± 1.73, respectively) (all P <0.05), while the budesonide intervention group compared with the control group, the difference was not statistically significant (P> 0.05). The linear correlation analysis of GITR / GITRL mRNA expression in each group showed that the expression of GITR / GITRL was positively correlated (R2 = 0.669, P <0.01). CONCLUSION: Neutrophils may be involved in the airway inflammatory process of asthma through the GITR / GITRL signaling pathway. Budesonide may reduce the airway inflammatory response in asthma by down-regulating the expression of GITR / GITRL on neutrophils.
