本文观察了氢化可的松对新生儿脐血单个核细胞(CBMC)产生γ-干扰素(IFN_γ)的影响。试检结果表明,药理浓度(10~(-6)M)氢化可的松对ConA 诱导新生儿CBMC产生IFN_γ有明显的抑制作用,其抑制幅度大于该浓度氢化可的松对成人单个核细胞产生IFN_γ的抑制作用。生理浓度(10~(-7)M)氢化可的松对成人单个核细胞生成IFN_γ不产生明显影响,却使新生儿CBMC产生IFN_γ的水平呈下降趋势。将甘草甜素(100μg/ml)与新生儿CBMC预孵育12小时,可部分地逆转药理浓度氢化可的松对新生儿CBMC产生IFN_γ的抑制作用。本文的结果表明新生儿CBMC对氢化可的松抑制IFN_γ产生的效应比成人敏感,提示在儿科临床使用糖皮质激素时,更应警惕其造成的机体免疫功能下降及引起易感的问题。而在使用糖皮质激素前先使用一定量的甘草甜素,有可能会缓解由使用糖皮质激素而引起的免疫抑制效应。
This article investigated the effects of hydrocortisone on the production of IFN-γ in neonatal cord blood mononuclear cells (CBMCs). The test results showed that the pharmacological concentration (10 ~ (-6) M) hydrocortisone significantly inhibited the IFN-γproduction of neonatal CBMC induced by ConA, and its inhibitory effect was greater than that of hydrocortisone on adult mononuclear cells IFN_γ inhibition. Physiological concentration (10 ~ (-7) M) hydrocortisone did not affect the production of IFN-γ by adult mononuclear cells, but decreased the level of IFN-γ produced by neonatal CBMC. Preincubation of glycyrrhizin (100 μg / ml) with neonatal CBMC for 12 hours partially reversed the inhibitory effect of pharmacological concentrations of cortisol on IFNγ production in neonatal CBMCs. The results of this paper show that neonatal CBMC on hydrocortisone IFN_γ production effect than adults sensitive, suggesting that the clinical use of glucocorticoid in pediatric patients should be wary of their immune function decline and cause the problem of susceptibility. The use of glucocorticoids before using a certain amount of glycyrrhizin, may be used to alleviate the use of glucocorticoid caused by immunosuppressive effects.
