目的 为缺血疗法在胰腺癌的临床应用提供基础研究资料。方法 在建立人胰腺癌细胞株裸小鼠胰腺原位移植瘤模型的基础上 ,结扎荷瘤裸鼠胃十二指肠动脉、胰十二指肠下动脉及背胰动脉 ,诱导胰腺右叶区域性缺血 ,观察缺血对裸小鼠胰腺移植癌生长、移植癌及癌周胰腺组织病理形态学的影响。结果 缺血组移植癌生长缓慢 ,倍增时间延长 ,移植癌体积、癌细胞增殖指数及蛋白含量在术后第 3、7、14天均显著低于对照组 (P <0 .0 1)。光镜下缺血组移植癌呈大片凝固坏死 ,周围有渐进性坏死细胞及炎症细胞浸润 ;癌周胰腺组织腺泡萎缩破坏 ,可见广泛纤维化及淋巴细胞浸润。结论 区域性缺血对裸小鼠胰腺移植癌有确切的杀伤和抑制效应 ,癌周胰腺组织的缺血性改变可能不利于移植癌的生长。
Objective To provide basic research data for the clinical application of ischemic therapy in pancreatic cancer. Methods The pancreatic adenocarcinoma xenografts of human pancreatic cancer cell lines were established on the basis of establishing a model of orthotopic transplanted pancreatic tumors in nude mice. The gastric adenocarcinoma, pancreaticoduodenal and dorsal pancreatic artery of nude mice were ligated and the right lobe of pancreas was induced Effects of Ischemia and Ischemia on Pathological Morphology of Transplanted Pancreatic Tumor and Pancreatic Cancer in Nude Mice. Results In the ischemic group, the growth of transplanted cancer was slow and the doubling time was prolonged. The volume of tumor, the proliferation index of cancer cells and the content of protein in the ischemic group were significantly lower than those in the control group on the 3rd, 7th and 14th days after operation (P <0.01). Under light microscopy, the transplanted carcinoma of the ischemic group showed large coagulation and necrosis with progressive necrotic cells and infiltration of inflammatory cells around the peri-pancreatic tissue. The acinar atrophy and destruction of pancreatic tissue around the peri-week showed extensive fibrosis and lymphocyte infiltration. Conclusions Regional ischemia has the exact killing and inhibiting effects on pancreatic cancer in nude mice. The ischemic changes of peri-pancreatic tissues may not be beneficial to the growth of transplanted carcinoma.
