研究野生型 p5 3基因对染石英尘肺泡巨噬细胞 (AM)培养上清导致的成纤维细胞非正常增殖和功能的影响。将体内、体外染尘 AM上清加入人胚肺成纤维细胞 (2 BS)中培养 2 4小时后 ,应用脂质体介导转染法将野生型 p5 3基因导入(2 BS)继续培养 2 4小时。结果 ,转染 p5 3基因组 2 BS细胞增殖和胶原合成较未转染组受到抑制。表明 p5 3基因对由于石英尘作用后的 AM培养上清所致 2 BS的非正常增殖和胶原合成有抑制作用 ,而对正常 2 BS无明显影响。
To investigate the effect of wild-type p5 3 gene on the abnormal proliferation and function of fibroblasts induced by cultured supernatant of dust-stained alveolar macrophages (AM). The in vivo and in vitro stained AM supernatants were cultured in human embryonic lung fibroblasts (2 BS) for 24 hours. Then the wild-type p5 3 gene was introduced into the culture medium (2 BS) by liposome-mediated transfection 4 hours. As a result, proliferation and collagen synthesis of 2 BS cells transfected with the p5 3 genome were suppressed as compared with the untransfected group. The results showed that p5 3 had no effect on the abnormal proliferation and collagen synthesis of 2 BS induced by AM culture supernatant after quartz dust, but had no obvious effect on normal 2 BS.
