目的鼻腔免疫小鼠,诱导生殖道局部黏膜的特异性免疫应答。方法将18只小鼠随机均分为三组:A组、B组小鼠鼻腔滴注人乳头瘤病毒(HPV)E7 10μg,B组静脉注射淋巴毒素(LT)0.05μg;C组用PBS作为对照。收集小鼠阴道冲洗液,ELISA测定HPV-E7特异性阴道IgA抗体滴度。在体内细胞毒性T淋巴细胞(CTL)活性分析实验中,用流式细胞仪检测靶细胞清除率。结果 A组阴道冲洗液中检测到HPV-E7特异性抗体滴度约为1∶20,高于B组的1∶40(P<0.05)。A组细胞杀伤率为(48.64±3.07)%,低于B组的(69.12±9.01)%(P<0.05)。结论HPV-E7经鼻腔免疫小鼠后,可以使生殖道局部黏膜产生特异性体液和细胞免疫应答;LT则有效地提高了生殖道局部黏膜免疫应答效应。
Objective To immunize mice with nasal mucosa to induce specific immune response to local mucosa of genital tract. Methods Eighteen mice were randomly divided into three groups: group A and group B, mice were injected intranasally with 10μg of HPV E7, group B injected with lymphotoxin (LT) 0.05μg, group C with PBS as Control. Mouse vaginal washings were collected and the HPV-E7-specific vaginal IgA antibody titers were determined by ELISA. In vivo cytotoxic T lymphocyte (CTL) activity assay, target cell clearance was measured by flow cytometry. Results The titer of HPV-E7-specific antibody in group A vaginal was about 1:20, which was higher than that in group B (P <0.05). The cell killing rate in group A was (48.64 ± 3.07)%, which was lower than that in group B (69.12 ± 9.01)% (P <0.05). Conclusion HPV-E7 immunized mice can produce specific humoral and cellular immune responses in the local mucosa of genital tract. LT can effectively improve the local mucosal immune response in genital tract.
