目的:研究尿激酶型纤溶酶原激活物(uPA)对高糖诱导系膜细胞(MC)增殖及α-肌动蛋白(α-SMA)表达的影响。方法:MC分组:对照组(5 mmol/L D-葡萄糖),高糖组(30 mmol/L D-葡萄糖)和uPA组(30 mmol/L D-葡萄糖+不同浓度uPA)。分别用四甲基偶氮唑盐比色法(MTT法)和流式细胞仪检测细胞增殖和细胞周期情况;激光共聚焦观察α-SMA表达。结果:高糖刺激MC增殖率明显增加,uPA呈剂量依赖性的阻止细胞由G0/G1期进入S期,抑制MC增殖。高糖增加α-SMA表达,随着uPA浓度增加,核周α-SMA表达较高糖组明显减少(P<0.01)。结论:uPA抑制高糖诱导的系膜细胞增殖和表型转化,对糖尿病肾小球硬化有一定的治疗作用。
AIM: To investigate the effects of urokinase-type plasminogen activator (uPA) on the proliferation and the expression of α-actin (α-SMA) induced by high glucose in mesangial cells. Methods: MC group: control group (5 mmol / L D-glucose), high glucose group (30 mmol / L D-glucose) and uPA group (30 mmol / L D-glucose + different concentration uPA). The cell proliferation and cell cycle were detected by MTT method and flow cytometry respectively. The expression of α-SMA was observed by confocal laser scanning microscope. Results: The proliferation rate of MC was significantly increased by high glucose. UPA dose-dependently prevented cells from entering S phase through G0 / G1 phase and inhibited the proliferation of MC. High glucose increased the expression of α-SMA. With the increase of uPA concentration, the expression of α-SMA in the nucleus was significantly decreased (P <0.01). CONCLUSION: uPA can inhibit the proliferation and phenotype of mesangial cells induced by high glucose, and has a certain therapeutic effect on diabetic glomerulosclerosis.
