为探究紫甘薯提取物(Purple sweet potato extracts,PSPE)对更年期大鼠下丘脑神经递质及氧化应激的影响,以11~12月龄雌性SD大鼠作为自然更年期模型,将其分为更年组、阳性对照组和PSPE低、中、高剂量组,分别连续8周饲喂基础饲料、大豆异黄酮和不同剂量的PSPE。另选取3~4月龄大鼠作为青年对照组。检测下丘脑组织中5-羟色胺(5-HT)和多巴胺(DA)含量、单胺氧化酶(MAO)活力,计算5-HT/DA;血清和肝组织中总超氧化物歧化酶(T-SOD)、过氧化氢酶(CAT)活力,丙二醛(MDA)含量。PSPE可以显著降低更年期大鼠5-HT含量(P<0.01),升高DA含量,使5-HT/DA比值降低接近青年组,显著降低MAO活力(P<0.01),不同程度提高血清及肝组织中T-SOD、CAT活力,降低MDA含量。紫甘薯提取物可以通过调节下丘脑神经递质含量、增强机体抗氧化能力改善神经系统紊乱缓解更年期综合症。
To investigate the effects of purple sweet potato extracts (PSPE) on hypothalamic neurotransmitter and oxidative stress in climacteric rats, 11-12-month-old female Sprague-Dawley rats were used as natural climacteric models and divided into more Year group, positive control group and PSPE low, middle and high dose group, fed with basic diet, soy isoflavone and different doses of PSPE for 8 weeks. Another 3 to 4-month-old rats as a youth control group. The content of 5-HT and DA and the activity of monoamine oxidase (MAO) in the hypothalamus were measured to calculate 5-HT / DA. The levels of total superoxide dismutase (T-SOD) Catalase (CAT) activity and malondialdehyde (MDA) content. PSPE could significantly reduce the content of 5-HT in climacteric rats (P <0.01), increase the content of DA and decrease the ratio of 5-HT / DA in the youth group, and significantly decrease the activity of MAO (P <0.01) T-SOD, CAT activity in tissue, reduce MDA content. Purple sweet potato extract can regulate hypothalamic neurotransmitter content and enhance the body’s antioxidant capacity to improve nervous system disorders and alleviate menopausal syndrome.
