以珠子参节、节间和越冬芽萌发后的叶、根为试材,研究了不同消毒方法、外植体、植物生长调节剂及培养条件等对珠子参愈伤组织诱导的影响,以期获得愈伤组织诱导的最佳方法。结果表明:最佳消毒方法是75%酒精消毒15s后,越冬芽、节和节间分别加0.2%氯化汞消毒5、15、15min;节间为愈伤组织诱导最适宜外植体;2,4-D 2.0mg/L+KT 0.1mg/L对节间诱导愈伤组织效果最好,诱导率为96.67%;以MS为基础培养基添加浓度为5g/L的琼脂较适宜愈伤组织的诱导;温度25℃、光周期24h/d、光照强度6 200lx的条件有利于愈伤组织的诱导。
