用SSR分子标记构建9个美洲黑杨×青杨派杂种无性系和3个父母本的指纹图谱并进行遗传差异性分析。研究表明,所筛选出的12对引物共扩增出94条清晰条带,每对引物平均扩增条带为7.8条,其中,多态性条带85条,多态性比率达到90.4%;各杨树无性系间遗传相似系数在0.44~0.80之间,平均相似系数为0.62,并利用多态性条带构建出12个杨树无性系的分子指纹图谱。通过系统聚类分析发现,杂种无性系与母本美洲黑杨遗传差异性小,且在该指纹图谱中,每个无性系的谱带都不同,说明不同供试无性系在DNA分子水平上表现出一定的遗传差异,据此可将它们区别开来。
The fingerprints of nine Populus × Populus × Populus euphratica clones and three parents were constructed by SSR markers and their genetic differences were analyzed. The results showed that a total of 94 clear bands were amplified by 12 pairs of primers, and the average number of amplified bands per primer pair was 7.8. Among them, 85 bands were polymorphic with a polymorphism rate of 90.4%. The genetic similarity coefficients of poplar clones ranged from 0.44 to 0.80 with the average similarity coefficient of 0.62. The molecular fingerprints of 12 poplar clones were constructed using polymorphic bands. The results of phylogenetic tree analysis showed that there was little genetic difference between hybrid clones and female parent poplar, and the bands of each clone were different in this fingerprinting, indicating that the clones showed different DNA molecular levels A certain degree of genetic difference, according to which they can be distinguished.
