采用HPLC和GDAS分析测定G-CSF基因工程菌在发酵罐高密度发酵培养过程中菌体产生积累乙酸的含量,分析确定了乙酸含量与产物表达的量化关系,验证了乙酸抑制产物表达的作用。建立了快速测定菌体培养过程中乙酸积累含量检测和动态菌体产物表达量测定的方法。
The contents of acetic acid accumulated by G-CSF genetically engineered bacteria during high-density fermentation in fermentor were determined by HPLC and GDAS. The quantitative relationship between acetic acid content and product expression was analyzed, and the effect of acetic acid on the inhibition of product expression was verified. A rapid method for the determination of acetic acid accumulation and the determination of the amount of dynamic bacterial cells during the culture of mycelium was established.
