目的:观察丹龙醒脑方对老年肾虚血管性痴呆大鼠模型海马Bax,Bcl-2蛋白表达及神经元凋亡的影响。方法:用D-半乳糖注射和甲状腺片混悬液灌胃建立老年肾虚大鼠模型,随后行双侧颈总动脉间歇结扎加剪尾放血建立老年肾虚血管性痴呆大鼠模型。采用TUNEL法检测细胞凋亡,免疫组织化学方法检测Bcl-2、Bax蛋白表达。结果:与模型组比较,雌二醇组和丹龙醒脑方组抗凋亡基因Bcl-2蛋白表达显著性增加(P<0.01),凋亡基因Bax蛋白表达降低(P<0.01),凋亡细胞显著性减少(P<0.01)。结论:丹龙醒脑方能通过增强Bcl-2的表达,抑制Bax的表达,抑制神经元凋亡的方式,起到保护神经元,改善痴呆大鼠学习记忆能力的作用,具有和雌激素相似的效应。
Objective: To observe the effect of Danlongxingnao Prescription on the expression of Bax and Bcl-2 in hippocampus and the apoptosis of neurons in the aged model of renal vascular dementia. Methods: Aged rats with renal deficiency model were established by intragastric administration of D-galactose and thyroid gland suspension. The rat model of senile renal vascular dementia was established by intermittent ligation of common carotid artery and tail bleeding. Apoptosis was detected by TUNEL method and protein expression of Bcl-2 and Bax was detected by immunohistochemical method. Results: Compared with the model group, the expression of Bcl-2 protein in the estradiol group and the Danlongxingnaofang group was significantly increased (P <0.01), and the expression of Bax protein was decreased (P <0.01) The number of apoptotic cells decreased significantly (P <0.01). Conclusion: Danlongxingnao decoction can protect neurons and improve the learning and memory abilities of dementia rats by enhancing the expression of Bcl-2, inhibiting the expression of Bax and inhibiting the apoptosis of neurons.
