目的探讨消银解毒颗粒治疗银屑病的可能作用机制。方法以植物血凝素(PHA)和白细胞介素6(IL-6)共同诱导Jurkat细胞模拟银屑病T细胞异常增殖的病理模型,制备择取最优效药理血清浓度作用于该细胞模型,采用细胞活性试剂盒(CCK-8)法检测细胞活力,用流式细胞术检测细胞凋亡。结果当药理血清浓度为20%时,消银解毒颗粒组、凉血方组细胞抑制率显著高于解毒方组、阿维A组、蒸馏水组(P<0.01);消银解毒颗粒组细胞早期凋亡率最高,高于凉血方组、解毒方组、阿维A组、蒸馏水组(P<0.05或P<0.01)。结论消银解毒颗粒及其拆方凉血方、解毒方对银屑病T细胞异常增值的炎症病理模型均有抑制作用,且消银解毒颗粒和凉血方的抑制效果明显优于阿维A。
Objective To explore the possible mechanism of Xiaoyin Jiedu Granule in the treatment of psoriasis. Methods PHA and IL-6 together induced Jurkat cells to mimic the pathological model of abnormal proliferation of psoriatic T cells. The optimal concentration of pharmacological serum was selected for the cell model. Cell viability was detected by CCK-8 assay, and apoptosis was detected by flow cytometry. Results When the pharmacological serum concentration was 20%, the cell inhibitory rate of Xiaoyan Jiedu Granule group and Liangxuefang Granule group was significantly higher than that of Jiedu Fang group, Yawei A group and distilled water group (P <0.01) The apoptosis rate was the highest, higher than that in the group of Liangxuefang, Jiedu Fang, Avia A and distilled water (P <0.05 or P <0.01). Conclusion Xiaoyan Jiedu Granule and its demolition Fangxuexuefang and Jiedu Decoction have inhibitory effects on inflammatory pathological model of abnormal proliferation of T cell in psoriasis, and the inhibitory effect of Xiaoyin Jiedu Granule and Liangxue Fang is obviously better than that of Avi A .
