目的观察痹肿消汤(BZXD)对实验性关节炎大鼠血浆TNF-α和IL-1β的影响,探讨痹肿消汤治疗类风湿性关节炎的作用机制。方法75只SD大鼠随机分为4组,皮下注射Ⅱ型胶原诱导实验性关节炎模型,采用放免法检测各组大鼠不同时间血浆TNF-α和IL-1β水平。结果造模大鼠88%出现关节炎症状;造模25d后,模型组、痹肿消汤组及甲氨喋呤组TNF-α和IL-1β水平明显高于正常组(P<0.05);且模型组的TNF-α和IL-1β水平明显高于痹肿消汤组及甲氨喋呤组(P<0.01);随着时间的延长,模型组TNF-α和IL-1β水平逐渐升高,痹肿消汤组及甲氨喋呤组则逐渐降低;而痹肿消汤组TNF-α和IL-1β水平低于甲氨喋呤组(P<0.05)。结论TNF-α和IL-1β在RA滑膜组织炎症形成和发展中发挥着重要作用;痹肿消汤能下调血浆TNF-α和IL-1β的水平,其作用优于甲氨喋呤组。
Objective To observe the effect of BZXD on plasma TNF-α and IL-1β in experimental arthritis rats, and to explore the mechanism of BZXD in treating rheumatoid arthritis. Methods Seventy-five Sprague-Dawley rats were randomly divided into 4 groups. Type II collagen was injected subcutaneously to induce experimental arthritis. Radioimmunoassay was used to detect the levels of plasma TNF-α and IL-1β in rats at different times. Results Arthritis symptoms occurred in 88% of the model rats. After 25 days of modeling, the levels of TNF-α and IL-1β in the model group, Biaozhongxiao decoction group and methotrexate group were significantly higher than those in the normal group (P<0.05). The levels of TNF-α and IL-1β in the model group were significantly higher than those in the Biaozhongxiao Decoction group and Methotrexate group (P<0.01). As time went by, the levels of TNF-α and IL-1β in the model group gradually increased. The levels of TNF-α and IL-1β in Biaozhongxiao Decoction group and Methotrexate group were gradually lower than those in Methotrexate group (P<0.05). Conclusions TNF-α and IL-1β play an important role in the formation and development of inflammation in RA synovial tissue. BZNG can down-regulate the levels of plasma TNF-α and IL-1β, and its effect is better than that of methotrexate group.
