目的:探讨信号转导与转录激活因子3(STAT3)、MDM2基因在宫颈腺癌组织中的表达,并分析其在不同宫颈病变发生发展中的作用和意义。方法:选择2012年7月~2013年6月吉林省肿瘤医院收治的宫颈癌患者49例,宫颈癌组织为观察组,癌旁组织为对照组;选取同期良性病变正常宫颈组织28例作为正常组,采用免疫组化方法检测STAT3和MDM2蛋白在不同组织中的表达。结果:观察组宫颈癌组织STAT3和MDM2蛋白OD值分别为2.94±0.64和3.61±0.79,均高于对照组和正常组(P<0.05)。对照组与正常组比较差异有统计学意义(P<0.05)。Ⅲ级宫颈癌组织STAT3和MDM2蛋白OD值分别为3.18±0.92和4.70±0.83,均高于Ⅱ比较级和Ⅰ级各组间比较差异有统计学意义(P<0.05)。Ⅱ级与Ⅰ级比较,差异有统计学意义(P<0.05)。结论:宫颈癌组织STAT3和MDM2蛋白表达较强,正常组织较弱,且随着分级的增加而增强,宫颈癌的发生可能与STAT3和MDM2有关,随着STAT3和MDM2基因表达的增强,宫颈癌的发生率增加,病情加重。
Objective: To investigate the expression of signal transducers and activators of transcription 3 (STAT3) and MDM2 in cervical adenocarcinoma and to analyze their roles and significance in the development of different cervical lesions. Methods: Forty-nine patients with cervical cancer admitted to Cancer Hospital of Jilin Province from July 2012 to June 2013 were selected as the observation group and the adjacent non-cancerous tissue as the control group. Twenty-eight normal cervical tissues from benign lesions were selected as the normal group Immunohistochemistry was used to detect the expression of STAT3 and MDM2 protein in different tissues. Results: The OD values of STAT3 and MDM2 protein in the cervical cancer group were 2.94 ± 0.64 and 3.61 ± 0.79, respectively, which were significantly higher than those in the control group and the normal group (P <0.05). The difference between the control group and the normal group was statistically significant (P <0.05). The OD value of STAT3 and MDM2 protein in grade Ⅲ cervical cancer were 3.18 ± 0.92 and 4.70 ± 0.83, respectively, which were significantly higher than those in Ⅱ grade and Ⅰ grade (P <0.05). There was a significant difference between grade Ⅱ and grade Ⅰ (P <0.05). Conclusions: The expression of STAT3 and MDM2 in cervical cancer is strong, and the normal tissue is weak. With the increase of grade, the occurrence of cervical cancer may be related to STAT3 and MDM2. With the increase of STAT3 and MDM2 gene expression, cervical cancer The incidence increased, the condition worsened.
