目的 :探讨芥子气中毒早期肝组织内皮素_1(ET_1)、内皮素A型受体 (ETAR)的变化和意义。方法 :应用免疫组化及RT_PCR技术 ,观测大鼠芥子气中毒后肝组织ET的含量及ET_1,ETARmRNA水平的变化 ,以及该变化与肝组织损伤的关系。结果 :中毒后肝组织ET的含量先下降后增加 ,12h达峰值 ;中毒后肝preproET_1mRNA的表达在 2~ 6h低于对照组 ,12h增加达峰值 ;ETARmRNA的表达没有明显的变化。结论 :ET_1及其受体ETAR在芥子气中毒后肝的病理生理过程中可能起着重要作用。
Objective: To investigate the changes and significance of endothelin-1 (ET-1) and endothelin-type A receptor (ETAR) in early liver tissue of mustard gas poisoning. Methods: Immunohistochemistry and RT-PCR were used to observe the content of ET and the levels of ET 1 and ETARmRNA in rat liver after mustard gas poisoning, and the relationship between the changes and liver injury. Results: After poisoning, the content of ET in the liver tissue decreased first and then increased, reaching the peak at 12h. The expression of hepatic preproET_1mRNA in the liver tissue was lower than that in the control group at 2 ~ 6h after poisoning, and reached the peak at 12h. There was no significant change in ETARmRNA expression. Conclusion: ET_1 and its receptor ETAR may play an important role in the pathophysiology of liver after mustard gas poisoning.
