联合运用衰减全反射(attenuated total reflectance,ATR)探头与FTIR光谱仪,测定了新鲜离体的甲状腺癌转移性颈部淋巴结20枚和非转移性淋巴结69枚的FTIR光谱,比较两组光谱13个谱带的峰位及相对峰强等28个指标,找出甲状腺癌转移性淋巴结光谱的特征。与非转移性淋巴结相比,转移性淋巴结光谱中与蛋白、脂质、糖类、核酸相关的谱带均发生了明显的变化,具体如下:(1)与蛋白质相关的谱带的相对峰强I3280/I1460(P=0.029),I1640/I1460(P=0.019),I1546/I1460(P=0.004)均显著升高;(2)脂类相关的相对峰强I1743/I1460显著降低(P=0.026),而I1400/I1460则明显增高(P=0.001);(3)与糖类相关的1165与1120cm-1处峰位P1165(P=0.019),P1120(P=0.000)发生明显蓝移(向高波数移动),相对峰强I1165/I1460显著降低(P=0.006);(4)与核酸相关的1085cm-1处峰位P1085发生明显地红移(向低波数移动)(P=0.029);(5)其他指认未明的谱带的相对峰强I1303/I1460(P=0.008),I1303/I1240(P=0.039)均明显升高。与非转移性淋巴结光谱相比较,甲癌转移的淋巴结光谱在多个谱带的峰位或相对峰强方面存在显著的差异。FTIR可应用于术中甲状腺癌转移性淋巴结的探测。
FTIR spectra of 20 newly diagnosed thyroid carcinoma metastatic cervical lymph nodes and 69 non-metastatic lymph nodes were measured by using attenuated total reflectance (ATR) probe and FTIR spectrometer. Thirteen spectra were compared between the two groups With the peak position and the relative peak intensity of 28 indicators to find the characteristics of thyroid cancer metastatic lymph node spectrum. Compared with non-metastatic lymph nodes, the bands related to proteins, lipids, carbohydrates and nucleic acids in metastatic lymph node spectrum have been significantly changed, as follows: (1) Relative peak intensity of protein-related bands I3280 / I1460 (P = 0.029), I1640 / I1460 (P = 0.019), I1546 / I1460 (P = 0.004) were significantly increased; (2) Relative peak intensity I1743 / ), And I1400 / I1460 significantly increased (P = 0.001). (3) The positions of P1165 (P = 0.019) and P1120 (P = 0.000) at 1165 and 1120 cm- (P = 0.006); (4) P1085 at the peak of 1085cm-1 associated with nucleic acid was significantly red-shifted (shifted to low wave number) (P = 0.029); (5) The relative peak intensity I1303 / I1460 (P = 0.008) and I1303 / I1240 (P = 0.039) of other unknown bands were significantly increased. Compared with the non-metastatic lymph node spectrum, lymph node metastasis of the carcinoma has significant differences in peak positions or relative peak intensities of multiple bands. FTIR can be applied to detect intraoperative thyroid cancer metastatic lymph nodes.
