目的对白刺提取物进行毒理学安全性评价的实验研究,为白刺提取物的安全性提供理论依据,为其开发利用提供依据。方法急性毒性试验(Test of Maximum Tolerated Dose法):清洁级昆明种小鼠40只,随机分为实验组和对照组,每组20只,雌雄各半。实验组剂量为15.0 g/(kg·BW),对照组为蒸馏水,经口灌胃后,连续观察14 d,观察中毒表现及动物死亡情况。骨髓细胞微核试验:取股骨骨髓,检查骨髓嗜多染红细胞微核发生率。精子畸形试验:取附睾制片,查小鼠精子畸形率。沙门菌回复突变试验(Ames试验):采用平板掺入法;分加与不加S-9混合液两种。结果急性毒性试验(MTD法):雌性和雄性小鼠均未见明显的中毒症状及死亡,两种性别的小鼠的(MTD法)LD50均>15.0 g/(kg·BW);3项遗传毒性试验结果均为阴性。结论在本次实验条件下,白刺提取物急性毒性试验属无毒级;3项致突变表明白刺提取物无明显遗传毒性作用。
Objective To study the toxicological safety evaluation of Nitraria extract, provide the theoretical basis for the safety of Nitraria extract and provide the basis for its development and utilization. Methods Test of Maximum Tolerated Dose Method: Forty clean Kunming mice were randomly divided into experimental group and control group, with 20 mice in each group. The dose of experimental group was 15.0 g / (kg · BW), the control group was distilled water. After oral administration, the mice were continuously observed for 14 days and the poisoning performance and animal death were observed. Marrow cell micronucleus test: take femoral bone marrow, check the bone marrow polychromatic erythrocyte micronucleus incidence. Sperm deformity test: take epididymis film, check the mouse sperm deformity rate. Salmonella Mutation Mutation Test (Ames test): Adopt the plate incorporation method; Add and add S-9 mixture two kinds. Results Acute toxicity test (MTD method): No obvious poisoning symptoms and death were found in both female and male mice. The LD50 of both sexes was> 15.0 g / (kg · BW) in MTD method. Three genetic Toxicity test results were negative. Conclusions Under the experimental conditions, the acute toxicity test of Nitraria extract is non-toxic; three mutagens showed that there is no obvious genotoxic effect of Nitraria extract.
