目的:观察以疏肝健脾、和胃安神法组方的康泰复方对腹泻型肠易激综合征(D-IBS)大鼠结肠黏膜促肾上腺皮质激素释放激素(CRH)及CRH-R1mRNA表达的影响。方法:将育龄7天的新生SD大鼠100只随机分出10只为正常组,余90只利用幼鼠母子分离加直肠刺激法复制D-IBS内脏高敏感性大鼠模型,造模成功后按体重随机分为对照组、康泰胶囊高、中、低剂量组和西药组各10只。模型复制成功后分别给予生理盐水、康泰胶囊1.52、3.04、6.08g/kg、马来酸曲美布汀片0.1g/kg灌胃,每天1次,连续2周,治疗后再次评价其腹部收缩反射(AWR)阈值,并运用RT-PCR法检测大鼠结肠黏膜CRH及CRH-R1mRNA的表达。结果:康泰胶囊能够升高内脏高敏感性大鼠AWR阈值,降低结肠黏膜CRH-R1mRNA的表达。结论:康泰胶囊对D-IBS内脏高敏感性大鼠有较好的治疗作用,其作用机制可能与影响大鼠结肠黏膜CRH-R1mRNA表达相关。
OBJECTIVE: To observe the effects of Shu Tai Jian Shen Decoction on the colonic mucosa-induced corticotropin-releasing hormone (CRH) and CRH-R1 mRNA expression in rats with diarrhea-predominant irritable bowel syndrome (D-IBS) influences. Methods: Ten neonatal SD rats of 7-day-old age were randomly divided into normal group (n = 10), and the remaining 90 rats were subjected to D-IBS visceral hypersensitivity rat model by maternal separation and rectal stimulation. According to body weight randomly divided into control group, Kangtai capsule high, medium and low dose groups and Western medicine group of 10. After successful replication of the model, normal saline, Kangtai Capsule 1.52,3.04,6.08g / kg, trimebutine maleate tablets 0.1g / kg were given orally, once daily for 2 weeks. After the treatment, the abdominal contraction (AWR) threshold, and RT-PCR was used to detect the expression of CRH and CRH-R1mRNA in colonic mucosa of rats. Results: Kangtai Capsule increased the threshold of AWR in visceral hypersensitivity rats and decreased the expression of CRH-R1mRNA in colonic mucosa. CONCLUSION: Kangtai capsule has a good therapeutic effect on visceral hypersensitivity in D-IBS rats, and its mechanism may be related to the expression of CRH-R1mRNA in colonic mucosa of rats.
