目的:为了探讨中华眼镜蛇毒(NNAV)对荷人鼻咽癌(NPC)裸鼠血浆丙二醛(MDA)和超氧化物歧化酶(SOD)活性的影响。方法:采用比色法检测经腹腔注射低、中或高浓度NNAV(1μg/ml、5μg/ml、10μg/ml)的荷人NPC裸鼠血浆MDA和SOD活性。结果:荷瘤鼠血浆MDA(842nmol/ml)比正常裸鼠(392nmol/ml)明显升高,而荷瘤鼠血浆总SOD(T-SOD)、锰SOD(Mn-SOD)和铜锌SOD(CuZn-SOD)活性(5219Nu/ml,2031Nu/ml,3188Nu/ml)比正常裸鼠(12564Nu/ml,75.11Nu/ml,50.53Nu/ml)低;低、中和高浓度NNAV均可使荷瘤鼠血浆MDA降低或至正常范围(562nmol/ml,5.17nmol/ml,4.58nmol/ml),并可使荷瘤鼠T-SOD、Mn-SOD和CuZn-SOD有不同程度的升高,其中以高浓度组为佳(10888Nu/ml,7227Nu/ml,3661Nu/ml,)。结论:结果提示NNAV的抑瘤作用可能与其降低荷瘤鼠自由基水平和提高SOD活性有关。
Objective: To investigate the effect of Naja naja atra Venom (NNAV) on malondialdehyde (MDA) and superoxide dismutase (SOD) activity in nude mice bearing human nasopharyngeal carcinoma (NPC). Methods: Colorimetric assay was used to detect the plasma MDA and SOD activity in nude mice bearing human NPAV (1μg/ml, 5μg/ml, 10μg/ml) injected intraperitoneally at low, medium or high concentrations. Results: The plasma MDA (842nmol/ml) in tumor-bearing mice was significantly higher than that in normal nude mice (392nmol/ml), while the total SOD (T-SOD) and manganese-SOD (Mn-SOD) in plasma of tumor-bearing mice. Copper-zinc SOD (CuZn-SOD) activity (5219 Nu/ml, 2031 Nu/ml, 3188 Nu/ml) was higher than normal nude mice (12564 Nu/ml, 75.11 Nu/ml, 50.53 Nu/ml )Low; low, medium and high concentrations of NNAV can reduce the plasma MDA in tumor-bearing mice to normal range (562 nmol/ml, 5.17 nmol/ml, 4.58 nmol/ml), and can make tumor mice T - SOD, Mn-SOD, and CuZn-SOD increased in varying degrees, with the high concentration group being preferred (108 88 Nu/ml, 72 27 Nu/ml, 36 61 Nu/ml,). Conclusion: The results suggest that the anti-tumor effect of NNAV may be related to the decrease of free radical levels and increase of SOD activity in tumor-bearing mice.
