目的:研究减压内部沸腾法提取黄柏中盐酸小檗碱的最佳工艺参数。方法:以盐酸小檗碱得率为评价指标,采用单因素考察法,研究解吸液、提取压力、提取温度、提取时间等因素对提取效果的影响。并与乙醇回流提取法进行比较。结果:减压内部沸腾法提取黄柏中盐酸小檗碱的最佳工艺为:药材细粉加入55%的乙醇溶液60mL,解吸30min,加入60℃的水150mL,减压提取(真空度为0.06MPa,水浴温度60℃),提取时间10min,提取2次。结论:减压内部沸腾法提取黄柏中盐酸小檗碱提取效果优于乙醇回流提取法,具有快速、高效、低成本等优点。减压提取工艺稳定,可在生产中推广应用。
Objective: To study the optimal extraction process of berberine hydrochloride from Cortex Phellodendri by decompression internal boiling method. Methods: Taking the yield of berberine hydrochloride as evaluation index, single factor method was used to study the effect of desorption solution, extraction pressure, extraction temperature and extraction time on the extraction efficiency. And compared with ethanol reflux extraction method. Results: The optimal extraction process of berberine hydrochloride in Cortex Phellodendri by boiling under reduced pressure was as follows: 60 mL of 55% ethanol solution was added to the medicinal powder, desorbed for 30 min, 150 mL of water was added at 60 ℃, and extracted under reduced pressure (the degree of vacuum was 0.06 MPa , Water bath temperature 60 ℃), extraction time 10min, extracted twice. Conclusion: The extraction of berberine hydrochloride from Cortex Phellodendri by decompression internal boiling is superior to that of ethanol reflux extraction, which has the advantages of rapid, high efficiency and low cost. Decompression extraction process is stable, can be popularized and applied in production.
