采用硫酸铵沉淀,SephadexG-75、ConA-Sepharose4B、DEAE-纤维素32柱层析法,从肾移植病人尿中分离并纯化α_1-微球蛋白(α_1-MG)。用此蛋白免疫家兔、获得其抗血清。抗血清稀释度为1:1000-1:3500,亲和常数为3.45×10 ̄9l/ml,与β_2-MG,白蛋白、IgG的交叉反应小于0.003%。用氯胺T法对α_1-MG进行碘标记,同时建立了α_1-MG放免分析方法。经方法学鉴定,灵敏度为1.60ng/ml,回收率均值为112%,批内变异系数为2.5%-3.3%,批间变异系数为2.4%-3.8%。正常人尿α_1-MG含量为X±SD=5.86±4.50μg/ml(n=102)。
Α1-microglobulin (α_1-MG) was isolated and purified from the urine of kidney transplant patients by ammonium sulfate precipitation, Sephadex G-75, ConA-Sepharose 4B, DEAE-cellulose 32 column chromatography. Rabbits were immunized with this protein to obtain their antiserum. The antiserum dilution was 1: 1000-1: 3500 and the affinity constant was 3.45 × 10 ~ 9l / ml. The cross-reaction with β_2-MG, albumin and IgG was less than 0.003%. The chloramine T method was used to label α_1-MG for iodine. At the same time, the method of radio-immunity analysis of α_1-MG was established. According to the methodological identification, the sensitivity was 1.60ng / ml, the average recovery was 112%, the coefficient of variation within the batch was 2.5% -3.3%, and the coefficient of variation between the lots was 2.4% -3.8%.
