跌打通痹膏对兔膝骨关节软骨细胞MMP-1,MMP-3,MMP-13及COL-Ⅱ的mRNA表达的影响

目的:研究跌打通痹膏对兔膝骨关节炎关节软骨细胞MMP-1,MMP-3,MMP-13及COL-Ⅱ的mRNA表达的影响,探讨跌打通痹膏干预软骨基质降解的作用机制。方法:新西兰兔60只随机分成空白组、假手术组、模型组、跌打通痹膏组、骨通贴组,每组12只。空白组不做任何处理,模型组、跌打通痹膏组、骨通贴组采用改良Hulth造模法,假手术组打开关节腔后不做处理,直接缝合。正常组、假手术组不予处理,模型组、跌打通痹膏组、骨痛贴组分别用胶带、跌打通痹膏、骨痛贴膏贴敷治疗4周。术后第5周各组随机选择1只兔处死验证造模,进行膝关节软骨形态肉眼观察和切片光镜观察,检测关节软骨中MMP-1,MMP-3,MMP-13及COL-Ⅱ的mRNA表达。结果:1)膝关节软骨形态肉眼观察和切片光镜观察:模型组与空白组有差异,跌打通痹膏组、骨通贴组与模型组比较有差异,跌打通痹膏组与骨通贴组比较无差异。2)MMP-1,MMP-3,MMP-13和COL-Ⅱ的mRNA表达检测:模型组与空白组比较,MMP-1,MMP-3及MMP-13的mRNA表达显著提高,差异有统计学意义(P<0.01),COL-ⅡmRNA表达显著降低,差异有统计学意义(P<0.01);跌打通痹膏组、骨通贴组与模型组比较,MMP-1,MMP-3及MMP-13的mRNA表达显著降低,差异有统计学意义(P<0.01),COL-ⅡmRNA表达显著升高,差异有统计学意义(P<0.05);跌打通痹膏组与骨通贴组比较,MMP-1,MMP-3,MMP-13及COL-Ⅱ的mRNA表达差异无统计学意义(P>0.05)。结论:1)跌打通痹膏和骨通贴对KOA均有治疗作用,可延缓KOA的病程,且两者间差异无统计学意义。2)跌打通痹膏通过抑制MMP-1,MMP-3及MMP-13的mRNA表达,减缓COL-Ⅱ降解,从而有效保护膝骨关节炎模型兔关节软骨,延缓关节软骨退变。 Objective: To study the effect of “Ditaitongbi” cream on mRNA expression of MMP-1, MMP-3, MMP-13 and COL-Ⅱ in rabbit knee osteoarthritis articular chondrocytes, Methods: Sixty New Zealand white rabbits were randomly divided into blank group, sham operation group, model group, Ditan Tong Bi Paste group and Gu Tong Paste group, with 12 rats in each group. The blank group did not do any treatment, the model group, the drop hit Tong Bi plaster group, modified group Gulutu improved Hulth modeling method, sham operation group open the joint cavity without treatment, direct suture. The normal group and the sham operation group were not treated. The model group, Daitong Tongbiao cream group and Gutongtie group were treated with adhesive tape, Daitong Tongbi cream and Gundao plaster respectively for 4 weeks. At the fifth week after operation, rabbits were randomly selected to sacrifice rabbits for verification of the articular cartilage morphology. The articular cartilage was observed under the naked eye and sliced ​​by light microscope. The expressions of MMP-1, MMP-3, MMP-13 and COL-Ⅱ in articular cartilage mRNA expression. Results: 1) Macroscopic observation of the articular cartilage and observation by slicing light microscope: There were differences between the model group and the blank group. There was a difference between the model group and the model group. No difference between groups. 2) The mRNA expression of MMP-1, MMP-3, MMP-13 and COL-II: Compared with the blank group, the mRNA expressions of MMP-1, MMP-3 and MMP-13 in the model group were significantly increased (P <0.01), the expression of COL-ⅡmRNA was significantly decreased, the difference was statistically significant (P <0.01); compared with model group, (P <0.01), COL-ⅡmRNA expression was significantly increased, the difference was statistically significant (P <0.05) The mRNA expression of MMP-1, MMP-3, MMP-13 and COL-II were not significantly different (P> 0.05). Conclusion: 1) DaTongTongBiao and GuTongPie have therapeutic effect on KOA, which can delay the course of KOA, and the difference between them is not statistically significant. 2) Daitong Tongbi Paste can effectively prevent knee joint articular cartilage and delay the degeneration of articular cartilage by inhibiting the mRNA expression of MMP-1, MMP-3 and MMP-13 and slowing COL-II degradation.
