目的 检测 TGF- β1、Cycin E、P2 7在原发性胆囊癌中表达 ,研究它们的蛋白产物在原发性胆囊癌发生、发展过程中表达特点及相互关系。方法 应用免疫组化 S- P法检测 36例原发性胆囊癌中 TGF- β1、Cycin E、P2 7的表达 ,并以同期 2 0例慢性胆囊炎作对照。结果 1.TGF- β1、Cycin E基因蛋白产物阳性表达率显著高于其它相应良性对照组。P2 7蛋白表达率显著低于相应良性对照组。 2 .TGF- β1、Cycin E蛋白在有淋巴结或远处转移、Nevin - 期患者中明显增高于无转移、Nevin - 期的患者。P2 7在高中分化、无淋巴结或远处转移、Nevin - 期的患者中阳性表达率明显高于低分化、有转移、Nevin - 期患者。 3.TGF- β1与 Cyclin E、P2 7两者分别呈正、负相关。 4 .TGF- β1、Cycin E阳性患者生存率明显低于 TGF- β1、Cycin E阴性患者 ,P2 7阳性患者生存率高于 P2 7阴性患者。L ong- rank检验显示生存率差异都有显著性。结论 联合检查 TGF- β1、Cycin E、P2 7在原发性胆囊癌中表达有助于反映原发性胆囊癌生物学特性、为预后判断提供参考指标 ,并为干预性基因治疗提供实验依据。
Objective To detect the expression of TGF-β1, Cycin E and P2 7 in primary gallbladder carcinomas and to study the expression and correlation of their protein products in the pathogenesis and progression of primary gallbladder carcinomas. Methods Immunohistochemical S-P method was used to detect the expression of TGF-β1, Cycin E and P2 7 in 36 cases of primary gallbladder carcinoma, and 20 cases of chronic cholecystitis were used as controls. The positive rate of TGF-β1 and Cycin E gene protein products was significantly higher than that of other corresponding benign control group. P2 7 protein expression was significantly lower than the corresponding benign control group. The expression of TGF-β1, Cycin E protein in patients with lymph node or distant metastasis, Nevin - stage patients was significantly higher than those without metastasis, Nevin - phase patients. The positive rate of P2 7 was significantly higher in Nevin - stage patients than in poorly differentiated, metastatic Nevin - stage patients with P2 7 differentiated in high school without lymph node or distant metastasis. TGF-β1 and Cyclin E, P2 7, respectively, were positive and negative correlation. The survival rate of TGF-β1 and Cycin E positive patients was significantly lower than that of TGF-β1 and Cycin E negative patients, and the survival rate of P2 7 positive patients was higher than that of P2 7 negative patients. L ong-rank test showed significant differences in survival rates. Conclusion The combined examination of TGF-β1, Cycin E and P2 7 expressions in primary gallbladder carcinomas can help to reflect the biological characteristics of primary gallbladder carcinomas and provide a reference index for prognostic judgment and provide an experimental basis for interventional gene therapy.
