应用四川省农业科学院分析测试中心实验室建立的转基因玉米MON863品系特异实时定量PCR方法,测定含量为1%的转基因玉米MON863样品(CRM)中旁侧片段(品系特异片段)的含量,并从扩增反应、数据处理以及微量移液器等不确定度来源评定测量的不确定度。结果表明,uA=1.9×10-2,uB=9.3×10-4,uC=1.9×10-2,U95=0.04,测量结果为1.108%±0.04。MON863品系特异实时定量PCR方法检测结果的主要不确定度来自检验过程中的随机效应。
The content of flanking fragments (lineage-specific fragments) in transgenic corn MON863 sample (CRM) with a content of 1% was determined by using a specific real-time PCR method based on the MON863 strain of transgenic corn established by the Laboratory of Analytical and Testing Center of Sichuan Academy of Agricultural Sciences. Uncertainty sources such as response, data processing and micropipettes assess the uncertainty of the measurement. The results showed that uA = 1.9 × 10-2, uB = 9.3 × 10-4, uC = 1.9 × 10-2, U95 = 0.04, and the measurement was 1.108% ± 0.04. The main uncertainty of MON863 strain-specific real-time PCR assay results comes from random effects in the assay.
