用色差仪测定黄芩根皮及断面的颜色,采用HPLC法测定黄芩中的黄酮类成分,用UV法测定黄芩药材中的总黄酮,以考察黄芩药材颜色与有效成分含量的相关性。结果,色差仪测定黄芩颜色各色度值(△L~*、△a~*、△b~*和△E~*)的RSD均<3%。相关性分析结果表明表面△L~*值、△E~*值、断面△a~*值与各化学成分均无显著相关性:表面△a~*值和△b~*值与黄芩苷、汉黄芩苷、总黄酮含量呈正相关。断面△L~*值、△b~*值与黄芩苷、汉黄芩苷及总黄酮含量呈正相关,与千层纸素A含量呈负相关:断面△E~*值与黄芩苷、汉黄芩苷含量呈负相关,与汉黄芩素、黄芩素含量呈正相关。结果表明采用色差仪测定黄芩药材色泽的方法科学、可行,可为黄芩原药材的质量评价及分级提供参考。
The color of the root bark and section of Scutellaria baicalensis Georgi was determined by colorimeter. The flavonoids in Scutellaria baicalensis Georgi were determined by HPLC. The total flavonoids in Scutellariae Radix were determined by UV. The correlation between the color and the content of Scutellariae Radix was studied. As a result, the RSDs of colorimetric values (△ L ~ *, △ a ~ *, △ b ~ * and △ E ~ *) of Scutellaria baicalensis Georgi were all less than 3%. The results of correlation analysis showed that there was no significant correlation between △ L ~ * value, △ E ~ * value and △ a ~ * value of the surface and each chemical composition: △ a ~ * value and △ b ~ Chinese baicalin, total flavonoids content was positively correlated. The △ L ~ * values and △ b ~ * values of the cross sections were positively correlated with the contents of baicalin, baicalin and total flavonoids, and negatively correlated with the content of Melaleuca paper A: the △ E ~ Content was negatively correlated with wogonin, baicalein content was positively correlated. The results showed that the method of colorimetric determination of Scutellaria Radix scientifically feasible, can provide a reference for the quality evaluation and grading of Scutellaria baicalensis original herbs.
