目的研究P16、P38、P53和CyclinD1蛋白在食道癌中表达的意义,为食道癌早期诊断提供依据。方法选用河北医科大学外科手术切除治疗的食管癌存档蜡块为实验标本共123例,将标本制成4μm厚病理切片后进行HE染色,采用免疫组合化学法检测病理切片中P16、P38、P53和CyclinD1蛋白的表达量。收集、整理123例患者临床资料,应用SPSS软件对相关数据进行分析,研究P16、P38、P53和CyclinD1蛋白与临床病理因素的关系。结果 123例食管癌患者组织中P16、P38、P53和CyclinD1阳性分别为59、64、44和36例,阳性率分别为47.97%、52.03%、35.8%和29.3%。统计分析显示,P16、P38、P53和CyclinD1蛋白表达与瘤组织侵润深度及淋巴结转移有相关性(P<0.05或P<0.01),吸烟组P53蛋白表达与吸烟量有相关性(P<0.05);P38、P53和CyclinD1蛋白表达与饮酒量有相关性(P<0.05),P53和CyclinD1蛋白表达与癌远处转移量有相关性(P<0.05)。讨论P16、P38、P53和CyclinD1的表达与食道癌的发生密切相关,可用于食道癌早期诊断和辅助治疗。
Objective To study the expression of P16, P38, P53 and CyclinD1 in esophageal cancer and provide the basis for the early diagnosis of esophageal cancer. Methods A total of 123 specimens of esophageal cancer were collected from Hebei Medical University for surgical excision. The specimens were made into 4μm thick sections and then stained with HE. Immunohistochemistry was used to detect the expression of P16, P38, P53 and The expression of CyclinD1 protein. The clinical data of 123 patients were collected and analyzed. Relevant data were analyzed by SPSS software to study the relationship between P16, P38, P53 and CyclinD1 protein and clinicopathological factors. Results The positive rates of P16, P38, P53 and CyclinD1 in 59 esophageal cancer patients were 59, 64, 44 and 36, respectively. The positive rates were 47.97%, 52.03%, 35.8% and 29.3% respectively. Statistical analysis showed that the expression of P16, P38, P53 and CyclinD1 were correlated with the depth of invasion and lymph node metastasis (P <0.05 or P <0.01), and the correlation between the expression of P53 protein and smoking in smoking group (P <0.05) P38, P53 and CyclinD1 were correlated with alcohol consumption (P <0.05). There was a correlation between the expression of P53 and CyclinD1 protein and distant metastasis of cancer (P <0.05). Discussion The expression of P16, P38, P53 and CyclinD1 is closely related to the occurrence of esophageal cancer, which can be used for the early diagnosis and adjuvant treatment of esophageal cancer.
