目的探讨神经酰胺与颗粒细胞凋亡的关系。方法 以PMSG处理的雌性Wistar大鼠为模型,制备了颗粒细胞悬液。应用TUNEL法及流式细胞术检测了神经酰胺对离体培养颗粒细胞凋亡的影响。 结果 体外培养的颗粒细胞存在自发性凋亡,神经酰胺处理的颗粒细胞凋亡率明显高于对照组(P<0.05);流式细胞术分析可见增强的凋亡峰。结论神经酰胺可能作为一种重要的信息分子参与大鼠颗粒细胞凋亡。
Objective To investigate the relationship between ceramide and granulosa cell apoptosis. Methods PMSG-treated female Wistar rats were used as models to prepare granulocyte suspension. The effects of ceramide on the apoptosis of granulosa cells in vitro were detected by TUNEL and flow cytometry. Results There was spontaneous apoptosis of granulosa cells cultured in vitro, and the apoptosis rate of ceramide-treated granulosa cells was significantly higher than that of the control group (P <0.05). The enhanced apoptosis peak was observed by flow cytometry. Conclusion Ceramide may participate in the apoptosis of rat granulosa cells as an important information molecule.
