目的:探讨bFGF单克隆抗体联合顺铂对黑色素瘤B16细胞的抑制作用。方法:用CCK-8法检测bFGF单抗及联合顺铂后对黑色素瘤B16细胞的抑制作用。结果:3株稳定分泌bFGF的单抗,分别命名为1B2F3A3、4F2C9A10和1E8F11B11,亚类鉴定均为IgG1。经ProteinG纯化的单抗和顺铂对B16细胞的抑制作用呈剂量依赖效应,当单抗1B2F3A3、4F2C9A10和1E8F11B11浓度为100mg/L时,对B16细胞的抑制率分别为(17.9±1.0)%、(13.1±2.3)%和(14.2±1.5)%,P=0.000;顺铂1mg/L时,黑色素瘤B16细胞的抑制率分别为(34.9±5.1)%;当1B2F3A3(100mg/L)与顺铂(1mg/L)联合应用时,B16细胞的抑制率为(66.2±1.8)%,P=0.001。结论:3株bFGF单抗能体外抑制小鼠黑色素瘤B16细胞的增殖,与顺铂联合应用能明显提高两者抑制作用。抗体与化疗药物联合使用将为肿瘤治疗提供更为有效的方法,其机制有待进一步研究。
Objective: To investigate the inhibitory effect of bFGF monoclonal antibody combined with cisplatin on melanoma B16 cells. Methods: The inhibitory effect of bFGF monoclonal antibody and cisplatin on melanoma B16 cells was detected by CCK-8 assay. Results: Three monoclonal antibodies secreting bFGF were named as 1B2F3A3, 4F2C9A10 and 1E8F11B11, respectively. All the subclasses were identified as IgG1. The inhibitory effect of ProteinG purified monoclonal antibody and cisplatin on B16 cells in a dose-dependent manner, when the monoclonal antibody 1B2F3A3, 4F2C9A10 and 1E8F11B11 concentration of 100mg / L, the inhibition rate of B16 cells were (17.9 ± 1.0)%, (13.1 ± 2.3)% and (14.2 ± 1.5)%, P = 0.000 respectively. The inhibitory rates of B16 cells in melanoma were (34.9 ± 5.1)% when cisplatin was 1 mg / L, When combined with platinum (1 mg / L), the inhibition rate of B16 cells was (66.2 ± 1.8)%, P = 0.001. CONCLUSION: Three monoclonal antibodies against bFGF can inhibit the proliferation of mouse melanoma B16 cells in vitro. The combined application of cisplatin and cisplatin can significantly increase the inhibitory effect of both. The combination of antibodies and chemotherapeutic drugs will provide a more effective method for the treatment of cancer, and its mechanism needs further study.
