目的探讨大鼠自主吸入异氟烷对肺脏肿瘤坏死因子-α(TNF-α)表达的影响。方法2005年2月至12月北京大学人民医院将实验用Wistar大鼠随机分为空气对照组(C组)、氧气对照组(O2C组)、0·7%和1·5%异氟烷吸入组,O2C组及异氟烷组又分为吸入4h、8h、吸入8h停药2h亚组。采用分子生物学方法测定TNF-α浓度及其基因表达和肺组织髓过氧化物酶(MPO)含量。结果各组间MPO含量差异无统计学意义(P(0·05)。1·5%异氟烷组吸入4h和8h后支气管肺泡灌洗液(BALF)中TNF-α浓度及肺组织TNF-α信使核糖核酸(TNF-αmRNA)均高于C组(P<0·05),停药2h恢复。吸入异氟烷4h和8h时,1·5%异氟烷组BALF中TNF-α浓度和肺组织TNF-αmRNA表达高于O2C组(P<0·05),与0·7%异氟烷组相比差异无统计学意义(P(0·05)。结论吸入较高浓度的异氟烷可逆性的增加大鼠肺脏TNF-α表达,但不会造成中性粒细胞的聚集。
Objective To investigate the effects of isoflurane inhalation on tumor necrosis factor-α (TNF-α) expression in lungs of rats. Methods From February to December 2005, Peking University People’s Hospital randomly divided experimental Wistar rats into two groups: air control group (C group), oxygen control group (O2C group), 0.7% and 1.5% isoflurane Group, O2C group and isoflurane group was divided into inhalation 4h, 8h, inhalation 8h withdrawal 2h subgroup. The concentration of TNF-α and its gene expression and the content of myeloperoxidase (MPO) in lung tissue were determined by molecular biology method. Results There was no significant difference in MPO content between groups (P> 0.05). TNF-α in bronchoalveolar lavage fluid (BALF) and TNF-α in lung tissue were significantly increased in 1.5% isoflurane group 4h and 8h after inhalation α mRNA (TNF-αmRNA) were higher than those in group C (P <0.05), and recovered after 2h withdrawal.At 4h and 8h after inhalation of isoflurane, the concentration of TNF-α in BALF of 1.5% isoflurane And lung tissue TNF-αmRNA expression was higher than that of O2C group (P <0.05), and no significant difference compared with 0.7% isoflurane group (P <0.05) .Conclusion Inhalation of higher concentrations of different Hyaluronan reversibly increases TNF-α expression in the lungs of rats but does not cause neutrophil aggregation.
