探讨剔毒护肝方抗肝纤维化的主要作用机理。方法:分离正常大鼠肝星状细胞,并传代培养。用剔毒护肝方给正常大鼠灌胃,制备药物血清,温育培养细胞。用~3H-TdR掺入法和MTT比色法观察细胞增殖,ELISA法测定上清液中Ⅰ型胶原含量,貂肺上皮细胞生长抑制MTT法检测培养上清液中的转化生长因子β_1活性。结果:剔毒护肝方药物血清能显著抑制肝星状细胞增殖,并能抑制肝星状细胞生成Ⅰ型胶原及产生转化生长因子β_1。结论:剔毒护肝方能抑制肝星状细胞增殖和分泌转化生长因子β_1,减少胶原生成,从而减弱肝星状细胞的自分泌放大效应。这可能是剔毒护肝方抗肝纤维化作用的主要机制之一。
To explore the main mechanism of action of Tiaodu Hugan Decoction on hepatic fibrosis. Methods: Rat hepatic stellate cells were isolated and passaged. The rats were given intragastric administration of Tiaoduhuganfang to prepare drug serum and incubated in cultured cells. The cell proliferation was observed by ~3H-TdR incorporation method and MTT colorimetric assay. The content of type I collagen in the supernatant was determined by ELISA. The growth inhibition of silicosis epithelial cells was assayed by MTT assay to determine the activity of transforming growth factor-beta 1 in culture supernatant. RESULTS: The serum of Tiaodu Hugan Prescription could significantly inhibit the proliferation of hepatic stellate cells and inhibit the production of type I collagen and transforming growth factor beta 1 in hepatic stellate cells. Conclusion: Tiedu Hugan Decoction can inhibit the proliferation of hepatic stellate cells and secrete transforming growth factor β_1, reduce collagen production, and weaken the autocrine amplification effect of hepatic stellate cells. This may be one of the main mechanisms of anti-fibrosis effect of Tiaodu Hugan Decoction.
