目的研究树突细胞(DC)与NC I-H460细胞融合体诱导的抗肿瘤作用。方法(1)从人外周血单核细胞诱导DC并与NC I-H460细胞融合。设融合细胞(FC)组、冲击细胞(PC)组及T细胞(TC)组,分别以融合细胞活化的T细胞、抗原冲击DC活化的T细胞及未活化T细胞为效应细胞,用乳酸脱氢酶法测定3种效应细胞对NC I-H460细胞的杀伤作用。(2)皮下注射NC I-H460细胞制备荷瘤裸鼠,将18只荷瘤裸鼠随机分为FC组、PC组及TC组,每组6只,接种上述3种T细胞,比较3组裸鼠肿瘤体积和重量。结果(1)对NC I-H460细胞的杀伤率,FC组为43.54%,PC组26.57%,TC组3.25%,3组杀伤作用比较,差异有统计学意义(F=5.47,P<0.05)。(2)FC组肿瘤体积明显小于PC组和TC组。FC组肿瘤重量为(1 129±123)mg,PC组为(1 709±160)mg,TC组为(3 344±288)mg,3组差异有统计学意义(F=37.05,P<0.01)。结论DC与NC I-H460细胞融合体可有效诱导抗肿瘤免疫,作用优于细胞抗原冲击的DC。
Objective To study the antitumor effect induced by fusion of dendritic cells (DCs) and NC I-H460 cells. Method (1) DCs were induced from human peripheral blood mononuclear cells and fused with NC I-H460 cells. The fusion cells (FC) group, the impingement cells (PC) group and the T cell (TC) group were established respectively. T cells activated by fusion cells, T cells activated by antigen and activated T cells were used as effector cells. Killing effect of three kinds of effector cells on NC I-H460 cells by hydrogenase method. (2) NCI-H460 cells were injected subcutaneously into nude mice, and 18 nude mice bearing tumor were randomly divided into FC group, PC group and TC group, with 6 mice in each group. The above three T cells were inoculated and compared with 3 groups Tumor volume and weight in nude mice. Results The killing rate of NC I-H460 cells was 43.54% in FC group, 26.57% in PC group and 3.25% in TC group. The killing effect of NCI-H460 was statistically significant (F = 5.47, P <0.05) . (2) The tumor volume in FC group was significantly smaller than that in PC group and TC group. The tumor weight in FC group was (1 129 ± 123) mg, in PC group was (1709 ± 160) mg and in TC group was (3 344 ± 288) mg, the difference was statistically significant (P <0.01) ). Conclusion The fusion of DC with NC I-H460 cells can effectively induce anti-tumor immunity, which is better than that of cell antigen-challenged DCs.
