目的 :初步探讨Hp菌株cagA、picB基因表达与胃粘膜IL 8分泌 ,消化性溃疡发病的关系。方法 :行Hp分离培养 ;采用PCR扩增技术检测其cagA、picB基因表达 ,ELISA法检测胃窦粘膜IL 8表达水平。结果 :(1)cagA+/picB+菌株感染者胃窦粘膜IL 8表达水平显著高于cagA-/picB-菌株感染者〔(2 5 5 .12± 75 .0 1)pg/mg蛋白vs (147.6 8± 86 .2 1)pg/mg蛋白 ,P <0 .0 1〕 ;(2 )消化性溃疡患者中cagA+/picB+菌株感染率显著高于慢性胃炎患者 (80 .0 %vs 5 4.3% ,P <0 .0 1) ;结论 :(1)cagA+/picB+菌株与诱导胃粘膜IL 8分泌有关。 (2 )武汉地区cagA+/picB+菌株感染可能与消化性溃疡的发病有关。
OBJECTIVE: To investigate the relationship between cagA and picB gene expression in Hp strain and the secretion of IL-8 in gastric mucosa and the incidence of peptic ulcer. Methods: Hp was isolated and cultured. The expression of cagA and picB genes was detected by PCR and the expression of IL 8 was detected by ELISA. Results: (1) The expression of IL 8 in antral mucosa of cagA + / picB + strains was significantly higher than that in cagA- / picB- infected patients ((2 5 5 12 ± 75 0.1) pg / mg protein vs (147.6 8 (2) The infection rate of cagA + / picB + strains in patients with peptic ulcer was significantly higher than that in patients with chronic gastritis (80.0% vs 5 4.3%, P <0.01, P <0.01) <0 .0 1). Conclusion: (1) The cagA + / picB + strain is related to the induction of IL 8 secretion in gastric mucosa. (2) cagA + / picB + infection in Wuhan may be related to the pathogenesis of peptic ulcer.
