目的:探讨TGF-β2对人正常星形胶质细胞株HA1800增殖和迁移的作用。方法:体外培养HA1800细胞,培养基中加入外源性TGF-β2,分0 ng/ml、2 ng/ml和10 ng/ml组。Ed U标记检测HA1800细胞的增殖情况;流式细胞术检测细胞周期;划痕实验检测细胞迁移能力。结果:TGF-β2能明显增加Ed U阳性细胞的数量,并增加S期和G_2/M期细胞的比例;外源性的TGF-β2还可以明显提升HA1800细胞的迁移能力。结论:TGF-β2能够调节体外培养的HA1800细胞的生长状态,促进HA1800细胞的增殖和迁移能力。
Objective: To investigate the effect of TGF-β2 on the proliferation and migration of human normal astrocyte cell line HA1800. Methods: HA1800 cells were cultured in vitro. Exogenous TGF-β2 was added to culture medium and divided into groups of 0 ng / ml, 2 ng / ml and 10 ng / ml. Ed U-labeled HA1800 cells were detected proliferation; flow cytometry cell cycle; scratch test to detect cell migration. Results: TGF-β2 significantly increased the number of Ed U-positive cells and increased the proportion of S phase and G 2 / M phase cells. Exogenous TGF-β2 also significantly increased the migration ability of HA1800 cells. Conclusion: TGF-β2 can regulate the growth of HA1800 cells in vitro and promote the proliferation and migration of HA1800 cells.
