目的探讨组织化学染色方法在骨肉瘤诊断中的应用。方法比较苦味酸天狼猩红染色法、改良丽春红三色法和Masson三色法在含有不同成熟度的成骨组织和胶原组织的骨肉瘤、骨旁骨肉瘤、骨膜骨肉瘤、骨外骨肉瘤、炎性肌纤维母细胞瘤、恶性纤维组织细胞瘤、软骨肉瘤、纤维组织增生伴骨化及钙化的染色规律。结果改良丽春红三色法显示,成骨组织染色呈橘红和蓝相间,均质;骨样基质到成熟骨的染色规律为橘红→淡蓝→深蓝;纤维组织呈蓝色条纹状,软骨组织不着色。天狼猩红染色法显示,成骨组织染色呈淡红、黄、橘红、深红相间,均质状;从骨样基质到成熟骨的染色规律为淡红→黄→橘红→深红→紫红;软骨组织呈淡红色,均质;纤维组织呈红黄相间条纹状。Masson三色法显示,骨样组织染色呈浅蓝色,成熟的骨小梁呈深蓝色;纤维组织呈蓝色条纹状。结论在骨肉瘤的诊断中,改良丽春红三色法和苦味酸天狼猩红染色法能较好的将骨样组织和不同成熟度的骨组织显现出来,优于Masson三色法,并可用于骨发育的研究中。
Objective To investigate the application of histochemical staining in the diagnosis of osteosarcoma. Methods Compared with picric acid Sirius red staining, modified Ponceau trichrome and Masson trichrome in osteosarcoma, periosteal osteosarcoma, osteosarcoma, osteosarcoma , Inflammatory myofibroblastic tumor, malignant fibrous histiocytoma, chondrosarcoma, fibrous tissue hyperplasia with ossification and calcification. RESULTS: The improved Ponceau red trichrome showed that the osteogenic tissue stained orange-red and blue-white, and homogenized. The staining pattern of bone-like matrix to mature bone was orange-red, light blue and dark blue. The fibrous tissue was blue streaks and the cartilage tissue No coloring Sirius red staining showed that the staining of osteogenic tissue was light red, yellow, orange red, dark red and white, homogeneous shape; from bone-like matrix to mature bone staining for the red → yellow → orange red → dark red → purple; Cartilage tissue was pale red, homogeneous; fibrous tissue was striped red and yellow. Masson’s trichrome method showed that the osteoid tissue was stained light blue, the mature trabecular bone was dark blue, and the fibrous tissue was blue streaks. Conclusions In the diagnosis of osteosarcoma, the improved Ponceau red color method and picric acid Sirius red color method can better show bone-like tissues and different maturity of bone tissue, which is better than Masson’s trichrome method and can be used In bone development research.
