目的研究亚砷酸(As_2O_3)对人肺腺癌A549的抑制和诱导凋亡作用及对C-myc基因表达的影响及可能机制。方法选用人肺腺癌A549细胞株,运用体外细胞培养法,MTT法,流式细胞术检测As_2O_3对人肺腺癌的抑制和诱导凋亡作用;用逆转录-聚合酶链反应(RT-PCR)方法检测C-myc mRNA的表达。结果 As_2O_3对人肺腺癌A549细胞具有抑制作用,其抑制率呈时间-剂量依赖关系。不同浓度的As_2O_3均可诱导凋亡。1.0μmol/L、2.0μmol/L的As_2O_3可下调C-myc的表达。结论 As_2O_3具有抗肿瘤作用,主要是通过诱导细胞凋亡实现的,其机制与下调C-myc表达有密切关系。
Objective To study the effects of arsenious acid (As_2O_3) on the inhibition of human lung adenocarcinoma A549 and the induction of apoptosis and its effect on C-myc gene expression and its possible mechanism. Methods A549 human lung adenocarcinoma cell line was selected. The inhibition and induction of As 2 O 3 on human lung adenocarcinoma cells were detected by MTT assay and flow cytometry (FCM). The expression of apoptosis was detected by reverse transcriptase-polymerase chain reaction ) Method was used to detect the expression of C-myc mRNA. Results As 2 O 3 inhibited human lung adenocarcinoma A549 cells in a time-and dose-dependent manner. Different concentrations of As 2 O 3 can induce apoptosis. 1.0μmol / L, 2.0μmol / L As_2O_3 can down-regulate the expression of C-myc. Conclusion As_2O_3 has anti-tumor effect, mainly through the induction of apoptosis, and its mechanism is closely related to the down-regulation of C-myc expression.
