目的:黏附分子的表达与动脉粥样硬化的发生发展有关。观察芪丹通脉片对实验性动脉粥样硬化大鼠外周血单个核细胞中细胞间黏附分子1和血管细胞黏附分子1mRNA表达的影响。方法:模型构建、样品采集与分析分别于2003-03/06,2003-06/09在解放军第四军医大学实验动物中心、西京医院检验科分生实验中心完成。将健康雄性SD大鼠72只随机分为6组:模型组、空白对照组、阳性对照辛伐他汀组、芪丹通脉片低剂量组、芪丹通脉片中剂量组、芪丹通脉片高剂量组,每组12只。采用高脂饮食配合口服维生素D3建立大鼠动脉粥样硬化模型,各组动物灌胃给药。采用半定量反转录聚合酶链反应的方法检测各组动物外周血单个核细胞中细胞间黏附分子1和血管细胞黏附分子1mRNA的表达,分析造模及各药物组细胞间黏附分子1和血管细胞黏附分子1mRNA表达的变化。结果:72只大鼠均纳入实验结果分析。细胞间黏附分子1和血管细胞黏附分子1基因的表达:模型组比空白对照组明显增加(1.37±0.08,1.40±0.06,P=0.000);辛伐他汀组及各中药组均明显低于模型组(P=0.000),且芪丹通脉片高剂量组的作用明显优于芪丹通脉片低剂量组(0.40±0.06,0.40±0.05;0.61±0.07,0.67±0.05,P=0.003,0.007)。结论:高脂饮食能使细胞间黏附分子1和血管细胞黏附分子1的表达明显增加,而芪丹通脉片各剂量组可以不同程度地下调血管壁内细胞间黏附分子1和血管细胞黏附分子1的表达。而且芪丹通脉片高剂量组的作用效果明显优于低剂量组。
Objective: The expression of adhesion molecules is related to the development of atherosclerosis. Observe the effect of Qidan Tongmai Tablet on the expression of intercellular adhesion molecule-1 and vascular cell adhesion molecule-1 mRNA in peripheral blood mononuclear cells of experimental atherosclerotic rats. Methods: Model construction, sample collection and analysis were performed at the Experimental Animal Center of the Fourth Military Medical University of Chinese PLA and the Laboratory of Clinical Laboratory Division of Xijing Hospital from March 2003 to June 2006 and from June 2003 to June 2003 respectively. 72 healthy male SD rats were randomly divided into 6 groups: model group, blank control group, positive control simvastatin group, Qidan Tongmai tablet low dose group, Qidan Tongmai tablet middle dose group, Qidantongmai Tablets of high-dose groups, 12 in each group. Semi-quantitative reverse transcription-polymerase chain reaction was used to detect the expression of intercellular adhesion molecule-1 and vascular cell adhesion molecule-1 mRNA in peripheral blood mononuclear cells of each group of animals, and to analyze the model and intercellular adhesion molecule-1 and blood vessel of each drug group.
