目的了解去势后不同时相雌性大鼠体内成骨细胞功能的变化特点,分析其机理。方法6个月龄清洁级健康雌性SD大鼠40只,随机均分为去卵巢组和假手术组,实验时间为4个月。分别于2、16周时每组各处死10只大鼠,取L4和血清,检测骨源性碱性磷酸酶(BALP)、骨钙素(BGP);透视电镜观察成骨细胞的分泌功能和电镜下的形态学变化。结果去势组大鼠体内BGP、BALP水平2周时皆升高,16周时与对照组差异无显著性意义。透视电镜下观察2周时成骨细胞功能活跃,16周时功能趋于静止。结论雌激素减少时,大鼠体内的成骨细胞可出现短暂的功能活跃,而后发生的成骨功能低下是导致骨质疏松症的一个因素。
Objective To investigate the changes of osteoblast function in female rats at different phases after castration, and to analyze its mechanism. Methods Forty clean healthy female SD rats of 6 months old were randomly divided into ovariectomized group and sham operation group. The experiment time was 4 months. At 2 and 16 weeks, 10 rats were sacrificed in each group. L4 and serum were taken for detection of bone alkaline phosphatase (BALP) and osteocalcin (BGP). Osteoblasts were observed under transmission electron microscope Morphological changes under electron microscopy. Results The levels of BGP and BALP in the castrate group were both increased at 2 weeks and no significant difference with the control group at 16 weeks. Osteoblasts were active at 2 weeks after the electron microscopy. The function tended to be static at 16 weeks. Conclusions When the estrogen is decreased, the osteoblasts in the rat may have transient functional activation, and the subsequent osteogenic dysfunction may be one of the factors leading to osteoporosis.
