灵芝多糖对癫痫大鼠大脑皮质和海马MAPK14、CaN和GSH-Px表达的影响

目的:观察灵芝多糖对癫痫大鼠大脑皮质和海马区丝裂原活化蛋白激酶14(MAPK14)、钙调神经磷酸酶(Ca N)和谷胱甘肽过氧化物酶(GSH-Px)的影响,进一步研究癫痫的发病机制及灵芝多糖的作用机制。方法:SD大鼠32只,随机分为正常对照组、模型组、灵芝多糖组和卡马西平组,每组8只。除正常对照组(用生理盐水腹腔注射)外,均采用戊四氮(PTZ)腹腔注射制作慢性点燃癫痫模型。在造模同时予以相应灌胃干预,灵芝多糖组予灵芝多糖150 mg/kg,1次/d,卡马西平组予卡马西平15 mg/kg的混悬液,2次/d,持续28 d;模型组和正常对照组予生理盐水灌胃。实验结束后断头取脑,采用免疫组化和比色法检测大鼠皮质和海马区MAPK14、Ca N和GSH-Px的表达。结果:模型组皮质和海马区MAPK14的表达较正常对照组明显增加,而Ca N和GSH-Px的活力明显低于正常对照组(P<0.01);灵芝多糖组和卡马西平组皮质和海马区MAPK14的表达较模型组明显降低,Ca N和GSH-Px的活力较模型组组明显增强(P<0.01)。结论:灵芝多糖能够降低癫痫大鼠脑中MAPK14的表达、增强Ca N和GSH-Px的活力,可能具有减轻癫痫发作、保护神经元的作用。 Objective: To observe the effects of Ganoderma lucidum polysaccharides on mitogen-activated protein kinase 14 (MAPK14), calcineurin (Ca N) and glutathione peroxidase (GSH-Px) in the cerebral cortex and hippocampus of epileptic rats , To further study the pathogenesis of epilepsy and the mechanism of Ganoderma lucidum polysaccharides. Methods: Thirty-two SD rats were randomly divided into normal control group, model group, Ganoderma lucidum polysaccharide group and carbamazepine group, with 8 rats in each group. Except normal control group (intraperitoneal injection with saline), chronic ignition epilepsy model was made by intraperitoneal injection of pentylenetetrazole (PTZ). Ganoderma lucidum polysaccharides were given 150 mg / kg once a day, and carbamazepine 15 mg / kg suspension twice a day for 28 days d; model group and normal control group were given saline. At the end of the experiment, the brain was decapitated. The expressions of MAPK14, Ca N and GSH-Px in cortex and hippocampus of rats were detected by immunohistochemistry and colorimetric method. Results: The expression of MAPK14 in cortex and hippocampus in model group was significantly higher than that in normal control group, while the activity of Ca N and GSH-Px in model group was significantly lower than that in normal control group (P <0.01); in ganoderic polysaccharide group and carbamazepine group, Compared with model group, the expression of MAPK14 in model group was significantly lower than that in model group (P <0.01). The activity of CaN and GSH-Px in model group was significantly higher than that in model group (P <0.01). CONCLUSION: Ganoderma lucidum polysaccharides can reduce the expression of MAPK14 and increase the activity of CaN and GSH-Px in the brain of epileptic rats, which may reduce the seizure and protect the neurons.
