目的建立测定卡马西平与茶碱血药浓度的反相高效液相色谱外标法。方法以二氯甲烷为萃取溶剂,采用外标法进行含量测定。色谱柱为XDB C18柱(150 mm×4.6 mm,5.0μm),流动相为甲醇-水(60∶40),流速为0.7 mL/min,检测波长为235 nm。结果卡马西平与茶碱的质量浓度线性范围分别为1.25~20μg/mL和2.5~40μg/mL,平均回收率分别为102.0%和101.4%,日内、日间精密度的RSD均小于5%。结论该法测定卡马西平与茶碱的血药浓度准确、简便、经济,适合于临床应用。
Objective To establish an RP-HPLC method for the determination of plasma concentrations of carbamazepine and theophylline. Methods Methylene chloride was used as extraction solvent, and the content was determined by external standard method. The column was XDB C18 (150 mm × 4.6 mm, 5.0 μm). The mobile phase was methanol-water (60:40) at a flow rate of 0.7 mL / min with a detection wavelength of 235 nm. Results The linear range of carbamazepine and theophylline was 1.25 ~ 20μg / mL and 2.5 ~ 40μg / mL respectively. The average recoveries were 102.0% and 101.4%, respectively. The RSDs of intracranial and intraday precision were less than 5%. Conclusion The method for the determination of carbamazepine and theophylline plasma concentration accuracy, simplicity, economy, suitable for clinical applications.
