应用简易实验室和层流架成功地繁育SPF级裸鼠,并建立起双标记—AFP和HBsAg~+的人肝癌裸鼠模型。移植瘤已传5代,生长稳定,潜伏期缩短;经组织病理学、超微结构病理学和免疫组织化学研究,确认移植瘤来自于人原发性肝癌。应用免疫试剂盒和电子显微镜技术进一步证实荷瘤小鼠移植瘤不仅分泌AFP,而且还分泌。HBsAg,瘤组织含有HBsAg蛋白纤丝样特征的细胞。一种适合于癌细胞存活的转运培养液初步用于肝癌的取材,更深入的工作有待于进一步开展。
SPF-like nude mice were successfully bred using simple laboratory and laminar flow cytometry and double-labeled AFP and HBsAg + human hepatocellular carcinoma nude mice models were established. Transplanted tumor has been passed 5 generations, stable growth, shorten the incubation period; histopathology, ultrastructural pathology and immunohistochemistry study confirmed the transplanted tumor from human primary liver cancer. Application of immune kits and electron microscopy further confirmed that tumor-bearing mouse xenografts not only secreted AFP but also secreted. HBsAg, a tumor cell containing fibrillar features of HBsAg protein. A suitable transport medium for the survival of cancer cells was initially used for the preparation of liver cancer, and further work needs to be further carried out.
