目的观察灯盏花素对脊髓损伤后大鼠运动功能恢复的影响,并初步探讨其相应机制。方法选取36只运动功能正常的健康成年大鼠,随机分为A、B、C三组,每组12只。Allen打击法(250g.mm)在T9处造成大鼠不完全脊髓损伤模型。造模成功后30min,A组腹腔注射甲泼尼龙琥珀酸钠(MP)30 mg/kg,B组腹腔注射等体积氯化钠注射液,C组腹腔注射灯盏花素(Bre)15 mg/kg。于术后1 d、3 d和7 d对各组进行后肢运动功能BBB评分、斜板试验、血清中丙二醛(MDA)和超氧化物歧化酶(SOD)测定。结果 A、C组BBB评分结果于术后1d和3d均高于B组(P<0.05),斜板试验测得的角度于术后1 d、3 d和7 d均高于B组(P<0.01)。术后1d和3d,A、C组SOD活性比B组明显增高(P<0.01);而于术后1d、3d和7d,A、C组MDA含量均明显低于B组。结论灯盏花素对脊髓损伤后大鼠运动功能具有较好的恢复作用。
Objective To observe the effect of breviscapine on recovery of motor function after spinal cord injury in rats and to explore its corresponding mechanism. Methods Thirty - six healthy adult rats with normal motor function were selected and randomly divided into three groups (A, B and C), with 12 rats in each group. Allen Strike (250g.mm) caused a model of incomplete spinal cord injury at T9 in rats. 30 minutes after model establishment, group A received intraperitoneal injection of methylprednisolone sodium succinate (MP) 30 mg / kg, group B received intraperitoneal injection of sodium chloride injection of equal volume, group C received intraperitoneal injection of breviscapine 15 mg / kg . The BBB score of hindlimb motor function, the slant plate test, the level of serum malondialdehyde (MDA) and superoxide dismutase (SOD) were measured at 1 d, 3 d and 7 d after operation. Results The BBB scores of groups A and C were higher than those of group B on the 1st and 3rd postoperative day (P <0.05). The angles measured by the oblique plate test were higher than those of the B group on the 1st, 3rd and 7th day after operation <0.01). At 1 and 3 days after operation, the activity of SOD in group A and group C was significantly higher than that in group B (P <0.01). At 1, 3 and 7 days after operation, the levels of MDA in group A and C were significantly lower than those in group B. Conclusion Breviscapine has a good recovery effect on motor function of rats after spinal cord injury.
