目的检测整合素β1、血管内皮生长因子(VEGF)及基质金属蛋白酶-9(MMP-9)在人膀胱移行细胞癌的表达,探讨整合素β1与膀胱移行细胞癌血管生成及浸润转移的关系。方法采用免疫组织化学SABC法检测59例膀胱移行细胞癌组织及15例正常膀胱黏膜组织中整合素β1、VEGF、MMP-9的表达。结果59例膀胱移行细胞癌中整合素β1、VEGF、MMP-9的阳性表达率分别为45.76%、64.41%和55.93%,15例正常膀胱黏膜组织中三者的阳性表达率分别为13.33%、6.67%和6.67%,膀胱移行细胞癌与正常膀胱黏膜组织间3种蛋白阳性表达率差异均有统计学意义(P<0.05,P<0.01,P<0.01);整合素β1、VEGF、MMP-9的表达与膀胱移行细胞癌的分期、分级相关;整合素β1、VEGF的表达与膀胱移行细胞癌的癌灶直径、单发/多发相关;整合素β1与膀胱移行细胞癌患者的年龄相关;整合素β1与VEGF及MMP-9之间的阳性表达均呈正相关。结论整合素β1在膀胱移行细胞癌中表达升高,并与分级、分期等临床病理指标相关,表明其具有促进膀胱移行细胞癌生长的作用。整合素β1可能通过协同VEGF及MMP-9作用促进膀胱移行细胞癌的血管生成及侵袭转移。
Objective To detect the expression of integrin β1, vascular endothelial growth factor (VEGF) and matrix metalloproteinase-9 (MMP-9) in human bladder transitional cell carcinoma and to investigate the relationship between integrin β1 and angiogenesis and invasion and metastasis in transitional cell carcinoma of the bladder. Methods Immunohistochemical SABC method was used to detect the expressions of integrin β1, VEGF and MMP-9 in 59 cases of bladder transitional cell carcinoma and 15 cases of normal bladder mucosa. Results The positive rates of integrin β1, VEGF and MMP-9 in bladder transitional cell carcinoma of 59 cases were 45.76%, 64.41% and 55.93%, respectively. The positive expression rates of the three in 15 cases of normal bladder mucosa were 13.33% 6.67% and 6.67%, respectively. There were significant differences in the positive rates of the three proteins between bladder transitional cell carcinoma and normal bladder mucosa (P <0.05, P <0.01, P <0.01) The expression of integrinβ1 and VEGF was correlated with the diameter of tumor site and single / multiple stage of transitional cell carcinoma of bladder. The expression of integrinβ1 was correlated with the age of bladder transitional cell carcinoma patients. Integrin β1 and VEGF and MMP-9 positive expression were positively correlated. Conclusion The expression of integrin β1 in transitional cell carcinoma of the bladder is increased, which is correlated with the clinicopathological parameters such as grading and staging. It indicates that integrin β1 has the effect of promoting the growth of bladder transitional cell carcinoma. Integrin β1 may promote the angiogenesis and invasion and metastasis of bladder transitional cell carcinoma through the cooperation of VEGF and MMP-9.
