通过 RT-PCR 和 RACE 的方法得到了编码‘津田’芜菁(Brassica rapa L. ssp. rapifera‘Tsuda’)BrCOP1 的 cDNA 序列,长 2 034 bp,编码 677 个氨基酸,与拟南芥同源性达 94%,命名为 BrCOP1,GeneBank收录号为 JF738111。Northern 杂交分析表明 BrCOP1 的表达没有组织特异性,而在整株幼苗及成熟植株肉质根表皮中受长波紫外线(UV-A)诱导表达,但成熟植株的直根表皮中表达量不随处理时间延长而增加。这暗示 BrCOP1 作为光控发育的重要调控因子,可能在 UV-A 诱导‘津田’芜菁花青素合成及光信号转导中起重要作用。
The cDNA sequence of BrCOP1 coding for ’Brassica rapa L. ssp. Rapifera’Tsuda’ was obtained by RT-PCR and RACE, which was 2 034 bp in length and encoded a protein of 677 amino acids with homology to Arabidopsis thaliana Up to 94%, named BrCOP1, GeneBank accession number JF738111. Northern blot analysis showed that the expression of BrCOP1 was not tissue-specific, but was induced by UV-A in the fleshy root epidermis of whole plants and mature plants, but the expression of BrCOP1 in the root epidermis of mature plants did not change with the treatment time increase. This suggests that BrCOP1, as an important regulator of photocontrol development, may play an important role in the UV-A induction of ’Tsuda’ turnip anthocyanin synthesis and optical signal transduction.
