目的:观察La(C7H5O3)2·(C9H6NO)对体外培养的Hela细胞的生长抑制作用,并进一步探讨其作用机制。方法:应用MTT法检测细胞增殖活性,流式细胞仪检测细胞周期分布,Annexin Ⅴ-FITC染色法检测细胞凋亡率。结果:不同浓度的La(C7H5O3)2.(C9H6NO)处理Hela细胞24~96h后,细胞增殖受到显著抑制,并呈浓度及时间依赖性;0.5、1.0、5.0、10.0μmol/L La(C7H5O3)2·(C9H6NO)处理72h后,Hela细胞G0/G1期细胞数量显著增加,S期细胞数量显著减少(P<0.01);各浓度组细胞凋亡率均显著增加(P<0.01)。结论:La(C7H5O3)2·(C9H6NO)对Hela细胞具有生长抑制作用,其机制与阻滞细胞周期及诱导凋亡有关。
OBJECTIVE: To observe the inhibitory effect of La (C7H5O3) 2 · (C9H6NO) on the growth of Hela cells in vitro and to explore its mechanism. Methods: MTT assay was used to detect cell proliferation. Cell cycle distribution was analyzed by flow cytometry. Apoptosis rate was detected by Annexin Ⅴ-FITC staining. Results: After treated with La (C7H5O3) 2. (C9H6NO) at different concentrations for 24-96 h, the proliferation of Hela cells was significantly inhibited in a concentration- and time-dependent manner. With 0.5, 1.0, 5.0 and 10.0 μmol / L La (C7H5O3) After treated with C9H6NO for 72h, the number of cells in G0 / G1 phase increased significantly and the number of cells in S phase decreased significantly (P <0.01). The apoptosis rate of Hela cells increased significantly (P <0.01). Conclusion: La (C7H5O3) 2 · (C9H6NO) can inhibit the growth of Hela cells. Its mechanism is related to arresting cell cycle and inducing apoptosis.
