目的:探讨化痰通络汤对大脑中动脉栓塞(MCAO)模型大鼠脑组织细胞间黏附分子(ICAM-1)、基质金属蛋白酶9(MMP9)表达的影响。方法:32只SD大鼠按体质量随机分为假手术组、模型组、尼莫地平组与化痰通络组。以大脑中动脉栓塞法建立MCAO大鼠模型,观察大鼠一般状况,Zea-longa神经功能评分,免疫组织化学染色法检测大鼠脑组织ICAM-1、MMP9表达。结果:与假手术组比较,模型组大鼠神经功能评分明显增高(P<0.01),脑组织ICAM-1、MMP9表达增强(P<0.01);与模型组比较,化痰通络组大鼠神经功能评分降低(P<0.05),脑组织ICAM-1、MMP9表达明显减弱(P<0.01)。结论:化痰通络汤可明显改善MCAO模型大鼠的神经功能和一般症状,其机制可能与抑制大鼠ICAM-1神经炎症标志物、MMP9脑组织损伤标志物的表达有关。
Objective: To investigate the effect of Huatan Tongluo Decoction on the expression of intercellular adhesion molecule (ICAM-1) and matrix metalloproteinase 9 (MMP9) in the rat brain of middle cerebral artery occlusion (MCAO) model. Methods: Thirty-two SD rats were randomly divided into sham operation group, model group, nimodipine group and Huatan Tongluo group according to their body weight. MCAO rat model was established by middle cerebral artery occlusion. The general condition of rats, Zea-longa neurological function score, and immunohistochemical staining were used to detect the expression of ICAM-1 and MMP9 in rat brain. RESULTS: Compared with the sham-operated group, the neurological scores of the model group were significantly higher (P<0.01), and the expression of ICAM-1 and MMP9 in the brain tissue was increased (P<0.01). Compared with the model group, the Huayu Tongluo group rats The score of neurological function was decreased (P<0.05), and the expression of ICAM-1 and MMP9 in brain tissue was significantly decreased (P<0.01). Conclusion: Huatan Tongluo Decoction can obviously improve the neurological function and general symptoms of MCAO rats. The mechanism may be related to the inhibition of the expression of ICAM-1 neuroinflammatory markers and MMP9 brain tissue damage markers in rats.
