本研究通过体外纯培养实验研究了3种胆汁酸盐:牛磺胆酸(TCA)、胆酸(CA)和脱氧胆酸(DCA),对沃氏嗜胆菌(Bilophila wadsworthia)菌株的生长状况的影响。比浊法和活菌计数法结果表明,TCA能显著促进该菌株生长,与添加相同摩尔浓度的牛磺酸(TAU)带来的效应接近,而CA和DCA对该菌株无显著作用。通过超高效液相色谱-质谱技术分析B.wadsworthia菌株对TCA的代谢的结果显示TCA可被该菌株分解产生CA和TAU。Ca2+沉淀法对B.wadsworthia菌株产胆盐水解酶的能力进行鉴定的结果表明该菌株具有产胆盐水解酶的能力。由此推断,TCA对B.wadsworthia菌株的促进作用主要是通过其水解产生的TAU,而非CA来发挥的。
In this study, three kinds of bile salts, ascorbic acid (TCA), cholic acid (CA) and deoxycholic acid (DCA), were studied in pure culture in vitro. The growth of Bilophila wadsworthia Impact. Turbidimetric method and viable count method showed that TCA could significantly promote the growth of this strain, which was similar to the effect of adding TAA with the same molar concentration, while CA and DCA had no significant effect on this strain. Analysis of the metabolism of TCA by B.wadsworthia strain by ultra performance liquid chromatography-mass spectrometry revealed that TCA can be decomposed by this strain to produce CA and TAU. The results of Ca2 + precipitation assay on the ability of B.wadsworthia to produce bile salt hydrolase show that the strain has the ability to produce bile salt hydrolase. It is concluded that the promotion of TCA to B.wadsworthia strain is mainly through the TAU produced by its hydrolysis, but not CA.
