目的:观察从日本粗榧果肉中分离得到的三种二萜类化合物18-hydroxyferruginol(A)、kayadiol(B)及hinokiol(C)对人白血病细胞K562和鼠肉瘤细胞S180生长的影响。方法:以人白血病细胞K562为研究对象,采用MTT法检测上述三种二萜类化合物对K562细胞生长的抑制率,用相差倒置显微镜观察细胞形态变化。根据检测结果,对三种化合物进行抗癌活性筛选,将筛选出的有效化合物kayadiol再作用于鼠肉瘤细胞S180,观察其对S180细胞的作用。结果:18-hydroxyferruginol及kayadiol对K562细胞有抑制其增殖的作用;其有效浓度分别是1×10-4mol/L和1×10-8mol/L~1×10-4mol/L,kayadiol对S180细胞也有一定的抑制作用。hinokiol对于K562细胞的增殖没有明显的抑制作用。结论:日本粗榧果肉中分离得到的二萜类化合物18-hydroxyferruginol(A)、kayadiol(B)对人白血病细胞K562的生长具有一定的抑制作用。
OBJECTIVE: To observe the effects of three diterpenoids, 18-hydroxyferruginol (A), kayadiol (B), and hinokiol (C), isolated from Japanese roughy pulp on the growth of human leukemia cell line K562 and mouse sarcoma cell S180. METHODS: Human leukemia cell line K562 was used as the research object. MTT assay was used to detect the inhibition rate of the above three kinds of diterpenoids on K562 cell growth. The morphology of the cells was observed by phase contrast inverted microscopy. According to the test results, the three compounds were screened for anti-cancer activity, and the selected compound kayadiol was further reacted with the mouse sarcoma cell S180 to observe its effect on S180 cells. Results: 18-hydroxyferruginol and kayadiol could inhibit the proliferation of K562 cells; the effective concentrations were 1×10-4 mol/L and 1×10-8 mol/L~1×10-4 mol/L, respectively, and kayadiol on S180 cells. There is also a certain degree of inhibition. Hinokiol has no obvious inhibitory effect on the proliferation of K562 cells. Conclusion: The diterpenoid compounds 18-hydroxyferruginol(A) and kayadiol(B) isolated from Japanese rough pod pulp have certain inhibitory effects on the growth of human leukemia cell K562.
